Starting item is really about personal preference and play. For a jungling Tsuku, your best bet is Assassin's Blessing. If you're intending to solo lane or are playing Clash/Duel, Attacker's Blessing is your go-to item. I then tend to rush Transcendence, unless I'm facing a quicker god. I'll jump into tier two boots if necessary on my first back to the fountian.
Buy as many mana/health potions as you deem necessary to outlast the enemy jungler/laner.
The first relic can be a tough choice, but I usually veer towards Aegis Amulet. Blink Rune can be used to initiate a fight and gank from the jungle (which is quite a tasty prospect, honestly).
"Oh no, two stacking items?! How ever will I build both?" you may ask yourself, but fear not! Tsukuyomi is currently over-powered and can easily clear minion waves and build both items. Having that bit of lifesteal is extremely helpful in teamfights and gives him enough staying power to outlast threats.
Arondight may be a surprising choice, but consider this: his Ultimate lands you directly ontop of an enemy of your choice. Landing and dealing an extra percentage of power when striking is VERY GOOD.
Of course, with Elixir of Speed online, you can swap Warrior Tabi for some other situational power item (or even defensive, honestly).
I find Brawler's to be the perfect solution for healing gods. Placing caltrops underneath gods actually procs this item effect, meaning they have reduced healing for 8 seconds while you begin your onslaught of attacks.
If you find yourself getting clapped, I might suggest Void Shield to replace Soul Eater or even Heartseeker, whatever the situation demands. This is also an alternative item when selling boots. If you're feeling like creating a bruiser Tsuku, Void Shield is a must. Berserker's is a notable item since they've improved it with the update, but I don't know if it's worth an item slot. Usually, when Tsuku is at low enough health, he will typically just perish.
If you don't like building stacks, are in a low scoring game, or generally dislike Soul Eater, Bloodforge is an alternative that provides great power and a bit of lifesteal.
You may be asking yourself, "Why is she ignoring the caltrops until the end? They add such a great debuff to the enemy!" Well, you're quite right. That slow is phenomenal, with a percentage of damage to boot. The simple answer is this: the rest of Tsuku's kit is just better. The caltrops are a great way to ensure the enemy will not leave your encounter alive, but I find that investing in Kusarigama and Dark Moon Shuriken earlier allows Tsuku to hit harder and stun longer earlier. I always improve the ultimate on any god as soon as I can, as that improves damage.

Tsukuyomi players need to have a plan when engaging and enemy. Just jumping in to lend a hand could actually be detrimental to your team if you don't think ahead about how to escape after securing a kill or two. Remember, as an assassin, your goal is to be a finisher. If you can confidently pick off a god, then do NOT HESITATE!
Typical engagements go as follows:
Tsukuyomi is definitely a forced to be reckoned with, even after his inevitable nerf. The entire kit is unique for an assassin, giving him ranged basics that are sure to finish off anyone limping away from a fight. Piercing Moonlight can launch you between enemies like a rocket, leaving behind death and destruction in your wake (and a high assist score). Dropping caltrops when enemies are pursuing or running allows you to gain ground, potentially saving your own life or ending another.
All in all, Tsukuyomi may not be the ninja we needed, but definitely the ninja we deserve.
